Думаю, имелось в виду, что праймеры не содержатся в мастермиксе, а добавляются вместе с матрицей перед каждым прогоном

Получается, да. Если речь именно об аликвотах.
Если речь просто о том, что праймеры надо добавлять каждый раз с матрицей, то никаких аликвот не надо

Не совсем понимаю, что за "sample" имеется в виду в первом предложении. Образец положительного контроля или исследуемый образец от пациента? Если от пациента, зачем его разбавлять?
